What is Difficulty?
Difficulty controls how challenging your server is. Peaceful has no hostile mobs, Easy is gentle, Normal is standard, and Hard is the toughest. Which you pick changes the whole feel of the server.
Changing Difficulty In-Game
Use the command in-game: /difficulty peaceful /difficulty easy /difficulty normal /difficulty hard
You can also use short forms like /difficulty p for peaceful. This needs operator permissions.

The in-game command resets on restart. For a permanent change, edit the config:
